报告摘要:

Multiphoton microscopy (MPM) is a powerful optical imaging technique in the biomedicine field by virtue of its subcellular resolution and biochemical specificity. Traditional MPM uses benchtop systems with free-space optics, which are bulky andnot portable, causing difficulties to be used in the clinic. In order to apply MPM for in vivo imaging in clinical applications, bench-top MPM microscopes need to be miniaturized into compact MPM endoscopes.A fiber-optic MPM micro-endoscope is desirable for clinical applications. We will introduce our work on developing fiber-optics based compact MPM system. The challengesin the fiber MPM system developmentwill be addressed, including fiber delivery of fs pulses, compact laser source, and probe design.

A multimodal imaging system that combines MPM with optical coherence tomography (OCT) will also be introduced. MPM and OCT each has its own advantages and limitations. By combining them into a single imaging platform, we can acquire MPM and OCT images from the same sample location. The OCT modality can be used to scan a large tissue area at high speed to find regions of abnormality. The MPM modality can be used to acquire high-resolution images to identify tissue diseases based on the morphology of cells and extracellular matrix. Multimodal imaging can be important for label-free imaging to obtain a sufficient set of parameters for reliable sample analysis. The system development and applications will be introduc
